Laborat\u00f3rium analytickej cytometrie Pr\u00edrodovedeckej fakulty UPJ\u0160 (LAC) pod veden\u00edm prof. RNDr. Petra Fedoro\u010dka, <\/a>CSc.<\/a> je vybaven\u00e9 \u0161pi\u010dkov\u00fdm konfok\u00e1lnym mikroskopom TCS SP5 X, ktor\u00fd pl\u00e1novite doplnil u\u017e existuj\u00facu infra\u0161trukt\u00faru tohto laborat\u00f3ria pozost\u00e1vaj\u00facu z invertovan\u00e9ho fluorescen\u010dn\u00e9ho mikroskopu DMI 6000 B vybaven\u00e9ho mikromanipul\u00e1torom a kultiva\u010dnou kom\u00f4rkou pre zber d\u00e1t zo \u017eiv\u00fdch buniek. Obidva mikroskopick\u00e9 syst\u00e9my dodala spolo\u010dnos\u0165 Leica Microsystems.<\/p>\n \u0160pi\u010dkov\u00e1 mikroskopick\u00e1 technika v celkovej hodnote viac ako 658 tis. \u20ac bola zak\u00fapen\u00e1 z prostriedkov projektu APVV VVCE-0001-07 \u2013 \u201eCentrum pre v\u00fdskum signal\u00f3mu\u201c a z projektu \u0160F \u201eSEPO I – Sie\u0165 excelentn\u00fdch pracov\u00edsk pre onkol\u00f3giu\u201c (ITMS 26220120024).<\/p>\n Konfok\u00e1lny mikroskop Leica TCS SP5 X<\/strong> je zariadenie ur\u010den\u00e9 na trojrozmern\u00fa mikroskopick\u00fa anal\u00fdzu a sledovanie dynamick\u00fdch zmien v \u010dase v bunk\u00e1ch a tkaniv\u00e1ch Intuit\u00edvne a jednoduch\u00e9 softv\u00e9rov\u00e9 rozhranie a plne automatizovan\u00fd syst\u00e9m umo\u017e\u0148uje \u013eahk\u00e9 zacvi\u010denie a obsluhu. Otv\u00e1raj\u00fa sa tak atrakt\u00edvne mo\u017enosti vyu\u017eitia t\u00fdchto syst\u00e9mov aj vo v\u00fdu\u010dbe \u0161tudentov a v ich zasv\u00e4covan\u00ed do sveta molekul\u00e1rnej cytol\u00f3gie.<\/p>\n V z\u00e1vere roka 2010 bolo toto laborat\u00f3rium vybaven\u00e9 \u010fal\u0161\u00edm \u0161pi\u010dkov\u00fdm pr\u00edstrojom \u2013 prietokov\u00fdm cytometrom CyFlow ML a prietokov\u00fdm cytometrom so sortrom BD FACSAria<\/strong> – z projektu \u201eBudovanie infra\u0161trukt\u00fary v centre excelentnosti SEPO-II\u201c (ITMS 26220120039) vr\u00e1tane obnovy \u010fal\u0161ej laborat\u00f3rnej techniky v celkovej hodnote skoro 750 tis. \u20ac.<\/p>\n Prietokov\u00fd cytometer\/sorter BD FACSAria je zariadenie umo\u017e\u0148uj\u00face multiparametrick\u00fa anal\u00fdzu fluorescencie r\u00f4znych \u010dast\u00edc (naj\u010dastej\u0161ie buniek) pripraven\u00fdch v suspenzii a ich n\u00e1sledn\u00e9 delenie na samostatn\u00e9 popul\u00e1cie na z\u00e1klade \u013eubovo\u013enej kombin\u00e1cie analyzovan\u00fdch parametrov. BD FACSAria umo\u017e\u0148uje analyzova\u0165 a triedi\u0165 r\u00fdchlos\u0165ou a\u017e 70 000 \u010dast\u00edc za sekundu. Takto je mo\u017en\u00e9 analyzova\u0165 a izolova\u0165 aj vz\u00e1cne sa vyskytuj\u00face elementy ako napr. kme\u0148ov\u00e9 bunky, n\u00e1dorov\u00e9 a nen\u00e1dorov\u00e9 bunky a pod.<\/p>\n Laborat\u00f3rium analytickej cytometrie je lokalizovan\u00e9 jednak v b\u00fdval\u00fdch zdevastovan\u00fdch skladov\u00fdch priestoroch \u00dastavu biologick\u00fdch a ekologick\u00fdch vied PF UPJ\u0160 v RBL pavil\u00f3ne a jednak v rekon\u0161truovan\u00fdch laborat\u00f3rnych priestoroch \u00fastavu na M\u00e1nesovej 23. Medzi \u0161tandardn\u00e9 aplik\u00e1cie patr\u00ed subcelul\u00e1rna lokaliz\u00e1cia r\u00f4znych prote\u00ednov, \u0161trukt\u00farne zmeny bunkov\u00fdch organel, zmeny fyziologick\u00fdch parametrov bunky ako s\u00fa pH, mitochondri\u00e1lny potenci\u00e1l, hladina Ca2+<\/sup> a pod. Z\u00e1rove\u0148 je mo\u017en\u00e9 vykon\u00e1va\u0165 aj sofistikovanej\u0161ie anal\u00fdzy vz\u00e1jomnej kolokaliz\u00e1cie \u0161trukt\u00far pomocou met\u00f3dy FRET (Fluorescence Resonance Energy Transfer).<\/p>\n
