Vybran\u00e9 flavoprote\u00edny a vhodne navrhnut\u00e9 mutanty bud\u00fa exprimovan\u00e9 v Escherichia coli <\/em>a purifikovan\u00e9 be\u017en\u00fdmi met\u00f3dami. Vytvoren\u00e9 prote\u00edny konjugujeme so \u0161pecifick\u00fdmi DARPin-mi (skupina navrhnut\u00fdch ankyr\u00ednov\u00fdch repetit\u00edvnych prote\u00ednov \u2013 prote\u00ednov\u00e1 skupina, ktor\u00e1 predstavuje anal\u00f3gy protil\u00e1tok), ktor\u00e9 \u0161pecificky rozpozn\u00e1vaj\u00fa receptory HER2. Tieto konjug\u00e1ty sa pou\u017eij\u00fa na bunkovej \u00farovni na demon\u0161tr\u00e1ciu ich \u00fa\u010dinnosti, tj. selekt\u00edvnej v\u00e4zby na bunky s receptormi HER2 a toxicity pripojen\u00fdch flavoprote\u00ednov vyvolanej svetlom. Pri dosiahnut\u00ed tohto cie\u013ea z\u00edskame n\u00e1stroj na vytvorenie generickej met\u00f3dy na transform\u00e1ciu v z\u00e1sade ak\u00fdchko\u013evek flavoprote\u00ednov na \u00fa\u010dinn\u00e9 fotosenzibiliz\u00e1tory.<\/p>\n\n\n\n

Konkr\u00e9tne, navrhovan\u00fd transportn\u00fd syst\u00e9m bude skon\u0161truovan\u00fd f\u00faziou molek\u00fal LDL (alebo HDL) so \u0161pecifick\u00fdmi DARPinmi, ktor\u00e9 maj\u00fa vysok\u00fa afinitu pre naviazanie na HER2 receptory nach\u00e1dzaj\u00face sa na povrchu n\u00e1dorov\u00fdch buniek mlie\u010dnej \u017e\u013eazy a taktie\u017e DARPinmi vyselektovan\u00fdmi pre v\u00e4zbu na prirodzen\u00fa a mutovan\u00fa formu EGFR, ktor\u00e9 s\u00fa lokalizovan\u00e9 na povrchu buniek p\u013e\u00facnych n\u00e1dorov. Bunky n\u00e1doru mlie\u010dnej \u017e\u013eazy boli vybran\u00e9 ako modelov\u00fd syst\u00e9m na valid\u00e1ciu \u00fa\u010dinnosti skon\u0161truovan\u00e9ho transportn\u00e9ho syst\u00e9mu DARLINK. Modul\u00e1rna \u0161trukt\u00fara DARLINKu umo\u017en\u00ed za\u010datie novej l\u00ednie biomedic\u00ednsky orientovan\u00e9ho v\u00fdskumu so zna\u010dn\u00fdm dopadom na oblas\u0165 cielenej terapie.
