anot\u00e1cia<\/u>:\u00a0Zne\u010distenie \u017eivotn\u00e9ho prostredia je jedn\u00fdm z najz\u00e1va\u017enej\u0161\u00edch probl\u00e9mov s\u00fa\u010dasnosti. Bioremedi\u00e1cia je pou\u017eitie \u017eiv\u00fdch organizmov (napr. bakt\u00e9ri\u00ed) na odstr\u00e1nenie kontaminantov, zne\u010dis\u0165uj\u00facich l\u00e1tok a tox\u00ednov zo \u017eivotn\u00e9ho prostredia, \u010dasto z p\u00f4dy a vody. Cie\u013eom pr\u00e1ce je identifik\u00e1cia halofiln\u00fdch bakteri\u00e1lnych kme\u0148ov, ktor\u00fdch enz\u00fdmov\u00e9 aktivity maj\u00fa potenci\u00e1lne vyu\u017eitie pri degrad\u00e1cii r\u00f4znych odoln\u00fdch polutantov (napr. azo farbiv\u00e1, r\u00f4zne organick\u00e9 molekuly), predstavuj\u00facich zelen\u00fa alternat\u00edvu pri obnove \u017eivotn\u00e9ho prostredia.<\/p>\n

anot\u00e1cia<\/u>: Celul\u00f3za je najhojnej\u0161ie sa vyskytuj\u00faci pr\u00edrodn\u00fd biopolym\u00e9r na Zemi a hlavn\u00e1 zlo\u017eka bunkovej steny rastl\u00edn. Ke\u010f\u017ee je to odoln\u00fd a vo vode nerozpustn\u00fd materi\u00e1l, \u010dasto kon\u010d\u00ed ako \u201eodpad\u201c z r\u00f4znych priemyseln\u00fdch procesov. Mnoh\u00e9 mikroorganizmy v\u0161ak disponuj\u00fa extracelul\u00e1rnymi enz\u00fdmami, tzv. celul\u00e1zami, ktor\u00e9 dok\u00e1\u017eu celul\u00f3zu \u010falej rozlo\u017ei\u0165 na gluk\u00f3zu, \u010do z nej rob\u00ed lacn\u00fd a obnovite\u013en\u00fd zdroj energie. Konverzia celul\u00f3zovej biomasy vyu\u017eit\u00edm celul\u00e1z je tak vysoko atrakt\u00edvny a potenci\u00e1lne udr\u017eate\u013en\u00fd \u201ezelen\u00fd\u201c pr\u00edstup k v\u00fdvoju nov\u00fdch bioprocesov a produktov. Z uveden\u00fdch d\u00f4vodov v \u00a0s\u00fa\u010dasnosti rastie z\u00e1ujem o h\u013eadanie nov\u00fdch bakteri\u00e1lnych kme\u0148ov, produkuj\u00facich celul\u00e1zy. Cie\u013eom bakal\u00e1rskej pr\u00e1ce bude pripravi\u0165 liter\u00e1rny preh\u013ead o celul\u00f3ze, celulolytick\u00fdch bakt\u00e9ri\u00e1ch, mikrobi\u00e1lnych celul\u00e1zach a ich biotechnologickom vyu\u017eit\u00ed. Praktick\u00e1 \u010das\u0165 pr\u00e1ce bude zameran\u00e1 na vyh\u013ead\u00e1vanie celulolytick\u00fdch bakt\u00e9ri\u00ed z extr\u00e9mnych (hypersalinn\u00fdch) prostred\u00ed, ktor\u00e9 \u010dasto produkuj\u00fa enz\u00fdmy s unik\u00e1tnymi vlastnos\u0165ami.<\/p>\n

anot\u00e1cia<\/u>: \u017divot pri vysok\u00fdch koncentr\u00e1ci\u00e1ch sol\u00ed je v pravom zmysle slova „extr\u00e9mny“ a prin\u00e1\u0161a mnoh\u00e9 v\u00fdzvy, ktor\u00e9 musia halofily – mikroorganizmy ob\u00fdvaj\u00face tieto prostredia \u2013 zvl\u00e1da\u0165. Ako s\u00fa\u010das\u0165 t\u00fdchto adapt\u00e1ci\u00ed produkuj\u00fa halofiln\u00e9 bakt\u00e9rie r\u00f4zne bioakt\u00edvne zl\u00fa\u010deniny, chemik\u00e1lie a enz\u00fdmy s jedine\u010dn\u00fdmi vlastnos\u0165ami, ktor\u00e9 s\u00fa hor\u00facim kandid\u00e1tom na poli biotechnol\u00f3gi\u00ed. Zvl\u00e1\u0161\u0165 atrakt\u00edvne s\u00fa halofiln\u00e9 enz\u00fdmy, ktor\u00e9 vykazuj\u00fa nezvy\u010dajn\u00e9 charakteristiky, najm\u00e4\u00a0vysok\u00fa stabilitu a aktivitu za extr\u00e9mnych podmienok (napr. funguj\u00fa vo vysoko slan\u00fdch roztokoch, v organick\u00fdch rozp\u00fa\u0161\u0165adl\u00e1ch \u010di pri extr\u00e9mnych hodnot\u00e1ch pH). Cie\u013eom bakal\u00e1rskej pr\u00e1ce bude spracova\u0165 teoretick\u00fd preh\u013ead o biotechnologickom vyu\u017eit\u00ed halofiln\u00fdch mikroorganizmov so zameran\u00edm na enz\u00fdmy, ako aj o \u0161trukt\u00fare halofiln\u00fdch prote\u00ednov a ich \u0161pecifik\u00e1ch. Praktick\u00e1 \u010das\u0165 pr\u00e1ce bude zameran\u00e1 na skr\u00edning bakt\u00e9ri\u00ed produkuj\u00facich extracelul\u00e1rne halofiln\u00e9 enz\u00fdmy (amyl\u00e1zy, prote\u00e1zy, lip\u00e1zy, nukle\u00e1zy, xylan\u00e1zy, at\u010f), ktor\u00e9 maj\u00fa potenci\u00e1lne biotechnologick\u00e9 vyu\u017eitie.<\/p>\n

4. Halofiln\u00e9 bakt\u00e9rie – bunkov\u00e9 tov\u00e1rne pre produkciu bioplastov<\/strong> –\u00a0VO\u013dN\u00c1<\/span><\/strong><\/h4>\n

anot\u00e1cia<\/u>: Bioplasty s\u00fa plasty produkovan\u00e9 mikroorganizmami, vykazuj\u00face vysok\u00fa biologick\u00fa odb\u00farate\u013enos\u0165 (daj\u00fa sa rozlo\u017ei\u0165 p\u00f4soben\u00edm mikr\u00f3bov) a biokompatibilitu. Tieto polym\u00e9ry pritiahli v posledn\u00fdch desa\u0165ro\u010diach zna\u010dn\u00fa pozornos\u0165, preto\u017ee s\u00fa ekologickou alternat\u00edvou plastov vyr\u00e1ban\u00fdch z petrochemick\u00fdch produktov a m\u00f4\u017eu by\u0165 navy\u0161e biosyntetizovan\u00e9 z obnovite\u013en\u00fdch zdrojov, ako s\u00fa cukry, rastlinn\u00e9 oleje, rastlinn\u00e1 biomasa \u010di dokonca oxid uhli\u010dit\u00fd. Pr\u00edkladom biologicky odb\u00farate\u013en\u00e9ho polym\u00e9ru s\u00fa tzv. polyhydroxyalkano\u00e1ty (PHA), produkovan\u00e9 \u010dasto bakt\u00e9riami a arche\u00f3nmi \u00a0hypersalinn\u00fdch prostred\u00ed (napr. morsk\u00e1 voda, slan\u00e9 pramene, so\u013eanky). Cie\u013eom bakal\u00e1rskej pr\u00e1ce bude \u00a0pripravi\u0165 liter\u00e1rny preh\u013ead o bioplastoch so zameran\u00edm na PHA, ich klasifik\u00e1cii, \u0161trukt\u00fare, sp\u00f4soboch skr\u00edningu producentov PHA, \u0161pecifick\u00fdch podmienkach potrebn\u00fdch pre produkciu t\u00fdchto bioplastov mikroorganizmami a met\u00f3dach ur\u010dovania ich chemickej \u0161trukt\u00fary. Experiment\u00e1lna \u010das\u0165 pr\u00e1ce bude zameran\u00e1 na vyh\u013ead\u00e1vanie halofiln\u00fdch bakt\u00e9ri\u00ed produkuj\u00facich PHA polym\u00e9ry, najm\u00e4 vyu\u017eit\u00edm detek\u010dn\u00fdch m\u00e9di\u00ed a na optimaliz\u00e1ciu produkcie PHA.\u00a0<\/p>\n

5. Kto de\u0161truje de\u0161truentov<\/strong>\u00a0– VO\u013dN\u00c1 <\/strong><\/span><\/h4>\n

anot\u00e1cia<\/strong><\/u>: drevo-rozkladaj\u00face huby s\u00fa hlavn\u00fdmi organizmami podie\u013eaj\u00facimi sa na de\u0161trukcii drevnej hmoty. Po odumret\u00ed plodn\u00edc sa v\u0161ak tieto st\u00e1vaj\u00fa prostred\u00edm pre existenciu \u010fal\u0161\u00edch de\u0161truentov. Cie\u013eom pr\u00e1ce je pripravi\u0165 liter\u00e1rny preh\u013ead o mikroorganizmoch (hub\u00e1ch a bakt\u00e9ri\u00e1ch) os\u00edd\u013euj\u00facich \u017eiv\u00e9 i odumret\u00e9 plodnice drevo-rozkladaj\u00facich h\u00fab a v praktickej \u010dasti sa zamera\u0165 na z\u00e1chyt a charakteriz\u00e1ciu popul\u00e1cie mikroorganizmov z tohto prostredia.<\/strong><\/span><\/p>\n
