\nZeta potenci\u00e1l je mierou ve\u013ekosti elektrostatick\u00e9ho odporu \/ pr\u00ed\u0165a\u017elivosti medzi \u010dasticami a je jedn\u00fdm zo z\u00e1kladn\u00fdch parametrov, o ktor\u00fdch je zn\u00e1me, \u017ee ovplyv\u0148uj\u00fa stabilitu koloid\u00e1lneho syst\u00e9mu. Meranie a kvantifik\u00e1cia zeta potenci\u00e1lu prin\u00e1\u0161a podrobn\u00fd poh\u013ead na pr\u00ed\u010diny polydisperzie, agreg\u00e1cie alebo vlo\u010dkovania napr. u monoklon\u00e1lnych protil\u00e1tok a vakc\u00edn. Z\u00edskan\u00e9 inform\u00e1cie m\u00f4\u017eeme pou\u017ei\u0165 na zlep\u0161enie formul\u00e1cie aj in\u00fdch disperzi\u00ed, emulzi\u00ed a suspenzi\u00ed. Meranie zeta potenci\u00e1lu je jedn\u00fdm zo sp\u00f4sobov ako skr\u00e1ti\u0165 testovanie koloid\u00e1lnej stability\u00a0 a to hlavne zn\u00ed\u017een\u00edm po\u010dtu vhodn\u00fdch formul\u00e1ci\u00ed, a t\u00fdm skr\u00e1ti\u0165 \u010das a n\u00e1klady na testovanie, ako aj pred\u013a\u017ei\u0165 trvanlivos\u0165 produktu.
\nV r\u00e1mci n\u00e1\u0161ho aktu\u00e1lneho v\u00fdskumu sme sledovali zeta potenci\u00e1l hydrof\u00f3bneho povrchu a jeho vplyv na adsorpciu dvoch terapeutick\u00fdch protil\u00e1tok IgG1 \uf06b-typu, herceptin a rituximab. Napriek tomu, \u017ee spom\u00ednan\u00e9 protil\u00e1tky s\u00fa sekven\u010dne ve\u013emi podobn\u00e9 (identita >80%), adsorpcia protil\u00e1tok v pr\u00edtomnosti excipientov prebieha odli\u0161ne. V pr\u00edpade rituximabu je pas\u00edvna adsorpcia na pevn\u00fd povrch ovplyvnen\u00e1 akumul\u00e1ciou vysoko polarizovate\u013en\u00fdch ani\u00f3nov v bezprostrednej bl\u00edzkosti protil\u00e1tky v d\u00f4sledku vz\u00e1jomn\u00fdch ne-elektostatick\u00fdch dispezn\u00fdch interakci\u00ed. Pri pr\u00edprave stabilnej formul\u00e1cie rituximabu odpor\u00fa\u010dame zv\u00e1\u017eenie resp. vynechanie excipientov s vysokou hodnotu polarizovate\u013enosti.
\nNa\u0161e v\u00fdsledky poukazuj\u00fa na potrebu individualizovanej kvantifik\u00e1cie zeta potenci\u00e1lu biofarmaceutick\u00fdch suspenzi\u00ed. Stanovenie a interpret\u00e1ciu zeta potenci\u00e1lu vieme uskuto\u010dni\u0165 v r\u00e1mci kontrahovanej slu\u017eby a\/alebo vedeckej spolupr\u00e1ce.<\/p>\n
